| 简介 | PA317细胞源自TK-NIH/3T3细胞，通过pBR322中的包装结构DNA(pPAM3)和pBR322中的单纯疱疹病毒胸苷激 酶基因共同转染构建而成。通过感染或转染将逆转录病毒载体导入这些细胞，结果生成可以感染许多哺乳动物细胞的 双嗜性载体颗粒。PA317细胞生成的病毒颗粒已成功地用于将基因导入人类，可能因为用于构建PA317细胞而转入的 DNA丢失，能够包装逆转录病毒载体的PA317细胞百分比随传代而减少。在含0.03mM次黄嘌呤、0.001mM氨甲 喋呤和0.02mM胸苷的培养基中短暂(大约5天)选择，可以选出保留了包装功能的细胞。选择后，PA317细胞应在HT 培养基(0.03mM次、0.02mM胸苷)中培养4天，以稀释残留的氨甲喋呤，此后PA317细胞不需选择可以稳定 至少1个月。 |
